It will probably take about 2 hours and 45 minutes to drive from Shannon Airport to Kinsale, provided you don't get caught in traffic. If you think you will be jet-lagged and very tired you should think of staying somewhere closer to your point of arrival.
I would choose a route that did not take you through Cork. It's a very confusing city to drive in with a high probability of getting lost. Maybe get off the N20 at Mallow and take the R619.

PS: Not sure I would want to do that on my first day, though it's possible. It's not quite as bad if you're from the east coast with only a five hour time difference.
Bill is right that trying to negotiate your way around Cork City is very confusing. There are many highways and ring roads. It is possible to get caught up in traffic that will make your journey much longer. So even though the smaller roads may seem like they are out of the way, you may actually make better time on them. But you must study your map in advance because signposting is not the best on the rural roads. Know the names of the towns and cities on your route so that you can head in the right direction.
