It still seems rather ambitious to me. It would be better to schedule the more abitious parts toward the middle or end of the trip - not at the begining.
It looks do-able, but you might have to re-think some of the 'Side Trips'.
Arriving in Dub at 6 AM after an overnight flight, heading to Cashel or Kilkenny is OK. Doing so, won't leave much time/energy for exploring both, though.
Likewise, on Day 6, Cashel to Killarney should be NO problem, but touring the Bearra along the way will make for a VERY long day. Day 7 is fine, but you'll do little more than sleep in Killarney.
Day 8 should be an easy day. Depending upon how much time you spend at the Folk Park, you may have time to add some other sites. There are some great music venues in Ennis.
Day 9 is a REALLY relaxed day. It would allow you to REALLY explore the Burren and the Clare coast.
Days 10 & 11 should allow for some exploration of Connemara.
Day 12 is fine, but will make for a very short Dublin visit.
Assuming these are all, 'Must Sees', were it ME:
I would visit Cashel en-route to Kilkenny and over-night in Kilkenny.
I would add an overnight stay in either Glengarrif, or Kenmare -- and Drop a night from Ennis.
I would drop off the car and THEN explore Dublin. Driving and parking in the City is NOT a great idea.

Looks doable and probably the pace I would opt for. You have your bases and they are about right so just see how the pace works.. I am a big fan of Connemara, wish I was in Clifden this weekend for the Trad Fest but not to be..That said not sure you have planned Dunguare Castle right as its on the way Too Galway along the Clare/Galway coastal drive.

I based my earlier comments on a mis-read. I see, NOW, that you were planning TWO nights, in Cashel -- NOt one.
Even WITH that, I would STILL recommend switching the lodging to Kilkenny, or at the very least, save touring Kilkenny -- as a Day Trip from Cashel, on the second day, to allow for Jet Lag.
I STILL have doubts about touring the Bearra and overnighting in Killarney, though.
